Margin is profit divided by selling price; markup is profit divided by cost. They're calculated from the same two numbers you enter but answer different questions — margin shows what share of the sale is profit, markup shows how much you added on top of cost.

Whatever you enter as cost is what's used — include shipping, packaging, or any other per-unit cost if you want the margin to reflect your true landed cost.
